Abstract

The present investigation was designed with the objective of evaluating the effects of different immersion cycles, characteristic of the embryos and composition of the liquid means on the regeneration of somatic embryos of Coffea arabica L. in bio- reactors of temporary immersion. It was carried out in the laboratory of biotechnology of the plants in JARC (Jimma Agricultural Research Centers, Ethiopia) between September of the 2006 and January of the 2008. Healthy leaves and with active growth starting from plants of coffee grown in hothouse were used as initial material. Hybrid F1 of Coffea arabica L. obtained of the crossing among wild agreements originated in Ethiopia, they were used during the experiment. Bio-reactors of temporary immersion (RITA) were used for the regeneration phase and development starting from embryonic suspensions. Different means, hormonal combinations, times and immersion cycles were proven in the regeneration and development of the embryos. Of all the evaluated treatments, the treatment 3 (1 minute of immersion every 12 hours and 300 mg of having added embryonic) it showed the biggest formation of embryonic mass for the three hybrid of evaluated coffee, of equal it forms it was this the treatment that bigger number of embryos in state ‘torpedo’ were obtained. 100 % was achieved from the regeneration to plant of the embryos with long cotyledons, 57 % for the medium ones and a 31 for the small ones. They were possible to regenerate the studied hybrid embryos of the three, to convert to plant and to acclimatize for this system.
